A CCR8 antibody and an ITGB6 ADC: A breakdown of BD value from mechanism to clinical practice
CCR8-targeted Treg clearing + ITGB6-targeted ADC,—— two differentiatedpipelines in a single biotech core judgment:
This asset portfolio includes two independent and complementary pipelines: one CCR8-targeted ADCC enhancement antibody that has entered clinical Phase Ib, and an ITGB6 targeted ADC in the IND-enabling-stage stage.
- CCR8 antibodies have completed the single-agent dose ramp-up and have shown safety signals and early efficacy in combination therapy;
- ITGB6 ADCs, leveraging conformation-selective antibodies and novel topoisomerase I inhibitor loadings, have demonstrated data comparable to world-leading molecules in preclinical models.
For buyers, the most valuable aspect is not purchasing all equity at once, but choosing an entry point based on their pipeline needs—the certainty of CCR8 antibody-anchored IO combination therapy, ITGB6 ADCs targeting differentiated ADC targets as strategic options, priced separately through regional licensing, phased exercise, or joint development.
Tumor immunotherapy is evolving from “single targets” to “combination therapies.” How to effectively eliminate immunosuppressive Treg in the tumor microenvironment and how to precisely target differentiated antigens on tumor cells are two core pathways.
Recently, two biotech pipelines came into our view: one is the CCR8 antibody HC006, which has entered clinical Phase Ib, and the other is the ITGB6 ADC HC029, which is in the IND-enabling-stage stage. These two pipelines respectively cover the two major hot tracks of IO and ADC, and both have clear differentiated design logic.
This article provides a logical breakdown and analysis of this project based on public information and does not constitute investment advice.
1. Differentiated Design and Early Clinical Validation
of HC006:CCR8 Antibody (1) Target Logic:
CCR8 is mainly expressed in regulatory T cells (Tregs) infiltrating tumors, with extremely low expression in normal tissues and peripheral blood Tregs. This “intratumor-specificity” makes it an ideal immuno-oncology target—by clearing immunosuppressive Treg within the tumor, releasing anti-tumor immune responses while avoiding the toxic risks associated with systemic immune activation. CCR8 also mediates the chemotactic transformation of Treg into the tumor microenvironment; blocking the CCR8 signaling pathway can simultaneously achieve the dual effect of “blocking Treg recruitment” and “clearing infiltrated Tregs.”
Globally, more than 10 CCR8-targeted drugs have entered clinical stages, but none have been approved for market launch. The track is still in the early stages of clinical validation, and there is a clear time window for differentiated assets.
(2) Product Design and Differentiation
HC006 introduces the S245D/A336L/I338E mutations into the IgG1 backbone to enhance ADCC and ADCP effects, efficiently clearing Treg cells within tumors through antibody-dependent cytotoxicity and phagocytosis. At the same time, CDC silencing mutations reduce complement-dependent cytotoxicity, thereby decreasing adverse reactions caused by nonspecific immune activation in peripheral normal tissues. This engineering approach of “enhancing effect functions while preserving the safety window” gives it a clear differentiated positioning among similar products.
Biochemical data showed that HC006 had a binding-level animolar affinity (EC50 of 0.346 nM and 0.252 nM) in human CCR8-overexpressing cell lines and endogenous CCR8-expressing cells Hut78, and cross-reactivity with NHP CCR8, supporting its convertibility in subsequent toxicological studies. Functionally, HC006 can effectively block the interaction between CCL1 and CCR8, suppress downstream signaling pathways, and prevent chemotactic peripheral Treg into the tumor microenvironment.
In parallel comparison with BMS 4A19 and Jounce/B16-AF analogs, HC006 demonstrated comparable or even superior in vitro activity in key functional indicators such as cell binding, receptor blockade, and ADCC/ADCP, indicating its foundational ability to compete globally.
(3) Clinical Progress and Preliminary Data
: HC006 has completed dose escalation as a single agent (18 subjects), with no dose-limiting toxicities (DLTs) reported in any dose group, indicating overall safety. As of January 2026, 12 subjects had been enrolled in the phase Ib combination therapy cohort, and no DLTs had been reported. In patients with solid tumors treated with after-line treatment, multiple tumor types have observed tumor shrinkage, and some clinical benefit signals begin to appear at lower doses—this observation is particularly important: for IO targets that rely on Treg clearance, seeing efficacy signals in the monotherapy phase means the mechanism is verifiable in humans.
(4) BD Value Assessment
As an important component of IO combination therapy, CCR8 antibodies have clear synergistic value for buyers with existing PD-1 positions. Global MNCs will continue to have a strategic need for next-generation IO portfolio assets amid the looming Keytruda patent cliff. HC006 has completed the dose ramp-up for monotherapy and entered the combination therapy cohort, placing it among the top domestic products in this target direction. For buyers, this is currently a suitable window to evaluate the CCR8 mechanism—the sector has yet to have a clear winner, but its safety has been largely verified. If subsequent combination therapy data are positive, asset valuations will see a significant leap.
| Evidence boundaries: Current data come from small samples and early clinical stages, which can support “safety and activity signals worthy of further verification,” but cannot be used to conclude that efficacy has been validated. Reading out data from future combination therapy cohorts will be a key point in determining whether this asset can enter registration development. |
2. Differentiated Design and Preclinical Validation
of HC029: ITGB6 ADC (1) Target Logic:
ITGB6 (integrin β6) is expressed very low in normal tissues but is significantly upregulated in various epithelial tumors, especially in non-small cell lung cancer (LUAD), It is highly expressed in head and neck squamous cell carcinoma (HNSCC), esophageal squamous cell carcinoma (ESCC), and pancreatic cancer (PAAD). Internal IHC data shows a LUAD positivity rate close to 98.5%, while both HNSCC and ESCC have positivity rates above 90%, with high expression (H-score 201-300) accounting for 20%-30%. ITGB6 expression is induced by the TGF-β pathway—elevated TGF-β levels in the tumor microenvironment directly drive upregulation of ITGB6, which in turn participates in TGF-β activation, forming positive feedback. This mechanism means ITGB6 is more expressed in “cold tumors,” which are precisely the populations with the lowest current immunotherapy response rates.
Seagen (now Pfizer)’s SGN-B6A is the fastest-progressing ADC asset for this target and has advanced to Phase II clinical trials. Although the Phase I data released by ASCO in 2025 did not meet market expectations, the biological logic behind the ITGB6 target continues to attract ongoing attention from the industry.
(2) Product Design and Differentiation HC029
identifies the “extended closure conformation” of ITGB6, which highly matches the ITGB6 conformation under TGF-β pathway activation。 The key value of this design lies in the fact that elevated TGF-β levels in tumor tissue drive a shift in ITGB6 conformation from “closed” to “open/extended,” with HC029 prioritizing activation of ITGB6 to achieve precise identification of tumor tissue. In vitro binding data showed that LAP (a natural ligand of ITGB6) enhanced the binding between HC029 and ITGB6, suggesting that HC029 is more likely to recognize ITGB6 in active conformations.
HC029 uses a novel topoisomerase I inhibitor loading (CPT class), which is active in both proliferative and non-proliferating cells compared to traditional microtubule inhibitors MMAE/MMAF and can produce bystander effects. After 21 days of incubation at 37°C in crab-eating monkey plasma, the DAR value changed very little, and its stability was better than Enhertu®.
In vitro killing experiments, HC029 had an IC50 of about 26.7 nM in the Calu3 tumor cell monoculture system, but in the CAF (tumor-associated fibroblast) and TGF-β co-culture system, the IC50 dropped to 0.39 nM, increasing its killing activity by about 70-fold. =
(3) BD Value Judgment
: Currently, only SGN-B6A is in Phase II clinical trials in the ITGB6 ADC sector, while second-tier assets are mostly in preclinical to IND stages. If HC029 successfully advances to IND, it will rank among the top two globally. For buyers, this is a reasonable window to evaluate ITGB6 assets—target biology has been preliminarily validated, but the track landscape has yet to solidify.
For buyers with existing ADC technology platforms or oncology pipelines, HC029 can serve as a strategic target for ITGB6. Clinical data from SGN-B6A provide a reference for target translation. HC029’s differentiated features in target selection (conformational recognition), payload design (CPT type), and plasma stability, if validated in clinical practice, will give it a clear clinical differentiated position in the ITGB6 track.
| Evidence boundaries: All current data come from preclinical models, and there is inherent uncertainty in the clinical translation of ADCs. However, the target already has clinical data from SGN-B6A as a reference, and ITGB6’s biological verifiability surpasses that of novel targets. |
3. The Valueof the Two Pipelines
From the buyer’s perspective, the value of these two pipelines goes beyond a simple sum of their independent assessments. What truly makes this asset group worth focusing on is the synergistic effect of three logical lines.
(1) Mechanisms complement
the mainstream model of tumor treatment is shifting from “breakthroughs in single drugs” to “combination coverage.” PD-1 inhibitors combined with chemotherapy, anti-angiogenic therapy, and ADC have become the first-line standard in multiple tumor types. CCR8 antibodies and ITGB6 ADCs cover two key nodes in the tumor immune cycle.
- CCR8 antibodies act on immunosuppressive cells (Tregs) in the tumor microenvironment, releasing immobilization of effector T cells by clearing Tregs;
- ITGB6 ADCs directly target and kill tumor cells, releasing tumor antigens and activating immune responses. There is a clear synergistic basis between the two mechanisms—ADCs kill antigens released by tumor cells, providing a “target” for the immune system activated by CCR8 antibodies; After CCR8 antibodies eliminate Treg, the cytotoxic activity of effector T cells is enhanced, further amplifying the anti-tumor effect of ADCs.
This synergistic logic has been preliminarily validated in preclinical studies: in the MC38 and EMT-6 animal models, the TGI of CCR8 antibodies combined with PD-1 inhibitors reached 89.2% and 94.2%, respectively, far exceeding monotherapy levels. Although data on the combination of ITGB6 ADCs and CCR8 antibodies have not yet been released, the combination of “ADC+IO” has clinical validation in multiple tumor types (such as Enhertu combined with Keytruda for exploration in HER2-positive breast cancer). If these two pipelines can be developed together by the same buyer, their synergistic value will far exceed the combined value of each pipeline’s independent assessment.
(2) Track Window Period Clarifying
CCR8 and ITGB6 Targets Share a Common Feature: The track has been preliminarily validated but the pattern remains undecided.
More than 10 CCR8 products worldwide have entered clinical trials, but none have been approved for market launch yet. Currently, most of the leading molecules are in clinical phase I/II, and there is no clear registration data available for readout. HC006 has completed the dose ramp-up as a monotherapy and entered the combination therapy cohort, placing it in the first tier among domestic CCR8 antibodies. This point in time means: the sector has not yet emerged with a clear winner, so buyers can position themselves within a relatively reasonable valuation window, rather than waiting for the third phase data to be read and pay a high premium.
The ITGB6 ADC track is even earlier. Currently, only SGN-B6A has entered Phase II clinical trials worldwide, while second-tier assets are mostly in preclinical to IND stages. HC029 has entered the IND-enabling, and if progresses smoothly, it will rank among the top two globally. For buyers seeking differentiated targets in the ADC field, ITGB6 is currently one of the few ADC targets not yet monopolized by MNC.
Both pipelines are in the time window of “having crossed key risk nodes but not yet entering the valuation premium stage.” For discerning buyers, this is the most suitable entry point.
(3) Trading flexibility
: The two pipelines can be authorized separately or traded in combination. Buyers can choose the most suitable participation method based on their pipeline layout needs:
- For buyers who already have PD-1/PD-L1 pipelines,CCR8 antibodies can serve as an important component of IO combination therapies, forming combination therapy combinations with existing products to extend product lifecycles.
- For buyers of existing ADC technology platforms or HER2/HER2-low pipelines, ITGB6 ADCs can serve as a targeted complement to ADC pipelines, differentiating the existing product portfolio.
- For buyers with both IO and ADC capabilities, combining the two pipelines offers greater strategic value—not only can they advance both pipelines separately, but there is also an opportunity to explore combination therapies combining CCR8 antibodies + ITGB6 ADCs, building differentiated combination advantages in the field of solid tumor treatment.
The flexibility of the transaction structure is also reflected in risk sharing. CCR8 antibodies are supported by clinical data, allowing buyers to price based on confirmed safety and early efficacy signals; ITGB6 ADCs are still in the preclinical stage, and buyers can reduce early payment risks through phased exercise and milestone payments. The different stage attributes of the two pipelines can precisely provide buyers with a differentiated risk-reward mix.
| Types of buyers | Best suited for the point of action | Recommended structure |
|---|---|---|
| Existing I/O drug companies with PD-1 deployments | CCR8 antibody | Regional license, as a joint partner for PD-1 |
| Pharmaceutical companies with existing ADC platforms/pipelines | ITGB6 ADC | Strategic investment + equity locking |
| Large pharmaceutical companies with IO+ADC capabilities | Combination of two pipelines | Combined licensing, exploring IO+ADC integration |
4. Conclusion
CCR8 antibodies and ITGB6 ADCs represent two typical categories of Chinese innovative drug assets with clear differentiation logic—one is IO target antibodies that have entered clinical validation stages, and the other is preclinical ADC assets with differentiated designs. Both pipelines are in the window period where the track has been preliminarily validated but the pattern has not yet been finalized.
For buyers with clinical development and portfolio capabilities, the greatest value of this asset group lies not in the peak sales expectations of a single pipeline, but in the portfolio premium generated when both pipelines advance in synergy. Both pipelines are in the time window of “having crossed key risk nodes but not yet entering the valuation premium stage.” For discerning buyers, this is the most suitable entry point.
