1、 Non atrial fibrillation arrhythmias can also increase the risk of dementia, with cumulative exposure showing a dose-response relationship

Recently, Wang Yunlong’s team from Capital Medical University and Yang Pingzhen’s team from Southern Medical University collaborated to publish their latest research in the top cardiovascular journal, the European Heart Journal (EHJ). For the first time, they systematically identified the independent impact of full spectrum arrhythmias on the occurrence of dementia, filling the gap in research on the association between non atrial fibrillation subtypes and cognitive impairment.

The global burden of dementia continues to rise, and previous research in this field has only clearly linked atrial fibrillation/flutter to an increased risk of dementia. There is insufficient research on the long-term cognitive effects of other common subtypes of arrhythmias, and there is a lack of direct pathological evidence at the level of brain structural imaging. This study included a large sample population and systematically evaluated the cumulative exposure of three common new onset arrhythmias (atrial fibrillation, bradycardia/conduction block, ventricular arrhythmia) and their association with the onset of different types of dementia. The biological association was analyzed by combining brain magnetic resonance imaging (MRI) data.

The research results show that three different subtypes of arrhythmia can independently increase the risk of developing major subtypes such as all-cause dementia and Alzheimer’s disease, and there is a clear dose-response relationship between cumulative exposure time and dementia risk. The longer the exposure time, the more significant the risk increase. Brain MRI analysis further confirms that exposure to arrhythmia is significantly correlated with neurodegenerative changes such as brain atrophy and gray matter volume loss, providing direct imaging support for the pathological association between the two.

This study extends the association between arrhythmia and dementia from atrial fibrillation to the entire spectrum, providing key evidence-based support for primary prevention of dementia and long-term stratified management of arrhythmia patients. It has important guiding significance for cardiovascular public health practice.

2、 Air Force Medical University team discovers GSDME promotes melanoma development

In recent years, the incidence rate of skin melanoma has increased year by year. The existing targeted therapy and immunotherapy still have clinical pain points such as primary/secondary drug resistance and high long-term recurrence rate. Exploring new disease driving mechanisms and intervenable targets is the core direction of research in the field. In May 2026, the team led by Li Chunying from the Air Force Medical University published important research results in the authoritative journal “Cell Death&Differentiation”, revealing for the first time the non pyroptosis dependent oncogenic function of the Gasdermin family protein GSDME, which refreshed the inherent understanding of the function of this family protein in the field.

The Gasdermin family proteins have traditionally been defined as pyroptosis effector molecules, and previous studies have suggested that they exert tumor suppressive effects. However, their non pyroptosis dependent functions, especially their nuclear regulatory activity, have long been unclear. This study confirms that GSDME can serve as a novel epigenetic regulatory factor, promoting malignant progression of melanoma by regulating downstream targets. Research has shown that GSDME is significantly upregulated in melanoma, and its high expression is driven by epigenetic low methylation in the promoter region; Functional experiments have confirmed that knocking out GSDME can significantly inhibit the in vivo growth and distant metastasis of melanoma, and this effect is not dependent on CD8+T cell-mediated anti-tumor immunity, clarifying its cancer cell autonomous pro cancer function.

Mechanistically, the nuclear localized GSDME can form a ternary complex with chromatin remodeling factor SMARCA5 and nuclear phosphoprotein NCL, specifically enhancing chromatin accessibility of the mitotic regulated kinase PLK4 promoter, thereby promoting transcriptional activation of PLK4. The GSDME-SSMARCA5/NCL-PLK4 regulatory axis has been validated in clinical patient samples and has clear prognostic potential. Research suggests that GSDME exhibits functional heterogeneity in different tumors, and its non apoptotic dependent function needs to be further explored. This discovery provides a new candidate direction for targeted diagnosis and treatment of melanoma.

3、 The team of China Pharmaceutical University developed a new hydrogel to reverse drug resistance in immunotherapy

On May 19, 2026, the team of Qiao Haishi from China Pharmaceutical University published the latest research in the international top journal PNAS, and developed injectable dual pathway activated hydrogel, which successfully realized the reversal of “cold tumor” and the overcoming of ICB resistance.

The team first identified the expression characteristics of NOD2 signaling in tumor associated macrophages (TAMs) through single-cell transcriptome analysis of clinical samples of colorectal cancer, and identified functional specific TAMs subpopulations with high NOD2 expression, providing clear cellular target evidence for targeted interventions. Previous studies have confirmed that NOD2 signaling can regulate anti-tumor immunity and ICB response, but there has been a lack of safe and effective delivery strategies for efficient and persistent activation of this pathway within tumors. Based on this, the team constructed manganese doped alginate brine gel carrier MHY@PBCG The encapsulated polyarginine modified BCG vaccine can achieve in situ sustained-release drug delivery in tumors: polyarginine modification can significantly improve the efficiency of intracellular uptake of BCG. Manganese ion can not only maintain the stability of the hydrogel network structure, but also act as a natural STING agonist to synergistically activate the pathway, and finally realize the synergetic activation of NOD2/STING dual pathways in TAM.

This strategy can effectively reprogram the pro tumor phenotype of TAM, transforming immunosuppressive “cold tumors” into inflammatory “hot tumors”, significantly restoring the sensitivity of drug-resistant tumors to PD-1 therapy, and providing an innovative biomaterial strategy with clinical translational potential for breaking the ICB resistance of solid tumors.

4、 Nature: New effects of alpha ketoglutarate: affecting carnitine synthesis, DNA repair, and tumor resistance

On May 27th, Katherine M. Aird’s team at the University of Pittsburgh published a groundbreaking study in the top tier journal Nature, revealing a novel mechanism by which α – ketoglutarate (α – KG) promotes DNA damage repair and drives tumor drug resistance by regulating carnitine synthesis.

This study constructed a CRISPR gene knockout screening library covering 64 types of α – KG dependent dioxygenases, and screened for the rate limiting enzyme TMLHE in the de novo carnitine synthesis pathway in cancer cells with complete HR function, confirming that it is a key essential gene for cancer cell survival after DNA damage. At the mechanistic level, the high abundance of α – KG in tumor cells serves as a coenzyme to maintain high catalytic activity of TMLHE and promote carnitine synthesis; Carnitine, as a key intracellular acyl carrier, can provide acetyl raw materials for histone acetylation modification. By reshaping chromatin conformation, it promotes the recruitment of HR repair proteins to DNA double strand break sites, achieving efficient damage repair.

Clinical cohort data confirms the clear clinical value of this pathway: high expression of tumor TMLHE and elevated levels of peripheral blood acetylcarnitine are significantly associated with shortened progression free survival in HR positive tumor patients after radiotherapy and chemotherapy. This study breaks the traditional understanding that alpha KG is only a basal metabolic intermediate and suggests that combination therapy targeting TMLHE or depleting alpha KG in tumors can effectively reverse chemoradiotherapy resistance in HR dominant tumors, providing a new target and clear clinical translation approach for the development of a new generation of anti-tumor combination therapies.

5、 Nat Biotechnol: CRISPR gene editing now has’ quality inspectors’, new platform SMArT makes cell therapy safer!

Recently, a joint team from the Telethon Gene Therapy Institute in San Rafael, Italy, published research in Nature Biotechnology, developing a screening platform SMArT based on artificial transactivators, providing a new controllable quality control scheme for precise editing of cells.

The core innovation of SMArT is to construct a temporary “AND gate” report screening circuit for conditional activation: this platform does not directly change the homologous repair efficiency of gene editing, but designs screening markers that are only activated when the target gene is accurately inserted into a predetermined genomic site. Through positive selection, it efficiently enriches and edits the correct hematopoietic stem cells, while completely eliminating unqualified cells carrying potential dangerous mutations and off target integration.

In preclinical models of humanized hematopoietic reconstruction, SMArT enriched edited hematopoietic stem cells can stably reconstruct functional human blood systems, with significantly improved editing purity and safety compared to non enriched populations. Unlike gene knockout strategies used in globally approved gene editing therapies such as Casgevy, SMArT fills the quality control gap in targeted integrated editing and is compatible with various existing and emerging genome engineering technologies. At present, this technology is still in the preclinical research stage, and its clinical safety and effectiveness need further verification. However, this platform provides a new idea for the production of safe and controllable gene edited cell products, which is expected to accelerate the clinical translation of the next generation of precision gene editing therapies.

6、 Huake Liu Li’s team found that oat β – glucan enhances the immunotherapy effect of colorectal cancer

Immune checkpoint inhibitors (ICIs) are the core treatment for advanced colorectal cancer, but primary/secondary drug resistance results in an overall response rate of less than 30%. Developing safe and accessible sensitization strategies is currently a research hotspot in the clinical oncology field. The gut microbiota is a key endogenous regulator of ICIs efficacy, and dietary interventions based on dietary fiber are highly anticipated for sensitization due to their low cost and high compliance. Recently, Liu Li’s team from Huazhong University of Science and Technology published a groundbreaking study in the top tier journal of Cell, Cell Host&Microbe, which for the first time elucidated the complete regulatory mechanism of oat β – glucan sensitization and anti-PD-1 therapy, providing a new direction for clinical intervention.

This study confirmed through multidimensional validation that daily supplementation of 5 grams of oat β – glucan can selectively enrich the beneficial intestinal symbiotic bacteria Clostridium perfringens, significantly increase the intestinal abundance of its characteristic metabolites butyric acid and indole-3-acetic acid (IPA), and reshape the anti-tumor immune microenvironment from upstream to downstream: butyric acid can effectively activate the antigen presentation function of dendritic cells, and butyric acid and IPA together enhance the killing function of tumor infiltrating CD8+effector T cells, ultimately synergistically amplifying the anti-tumor effect of anti-PD-1 therapy.

This study was validated in mouse transplant tumor models, colorectal cancer patient cohorts, and small-scale healthy population intervention trials, preliminarily confirming the safety of oat β – glucan intervention and its ability to enhance protective metabolites. The complete regulatory axis of “dietary fiber beneficial bacteria anti-tumor metabolites immune activation” was clarified, providing solid mechanistic support and preliminary clinical evidence for highly accessible colorectal cancer immunotherapy sensitization regimens.

7、 Shanghai Jiao Tong University team reveals the role of m6A and APA interaction in maintaining T cell rest

The maintenance of resting state of T cells is the core guarantee of immune homeostasis. Abnormal resting regulation can directly induce autoimmune diseases, immune deficiency and even tumorigenesis. However, the post transcriptional regulation mechanism of resting T cells has not been clarified for a long time. Previous studies have confirmed that RNA N6 methyladenosine (m6A) modification and variable polyadenylation (APA) are independently involved in regulating T cell function, but the direct interaction between the two and the mechanism of regulating T cell resting remain unclear. On May 13, 2026, the team led by Li Huabing from Shanghai Jiao Tong University published the latest research results in the internationally authoritative journal Science Advances, systematically analyzing the molecular mechanism of the interaction between m6A and APA to maintain T cell rest.

This study confirms that the m6A “writing enzyme” METTL3 can directly interact with the core APA regulatory factor NUDT21, jointly guiding the poly (A) site selection of target mRNA. In vivo functional experiments have shown that T cell specific knockout of Nudt21 can break the resting state of T cells, induce T cell overactivation and accelerate apoptosis, ultimately leading to peripheral T cell loss and significant impairment of the body’s antiviral immune function.

At the mechanistic level, METTL3 can specifically recruit NUDT21 to bind to the proximal poly (A) site of Rragd mRNA, producing a long 3’UTR with m6A modification that restricts the protein expression of Rragd; The absence of Nudt21 can lead to a shortened 3’UTR of Rragd mRNA, upregulation of protein expression, and overactivation of the mTOR signaling pathway, disrupting T cell homeostasis. This research fills the research gap of T cell resting multi-layer post transcriptional regulatory network, and provides a new theoretical target for the treatment of immune related diseases and the development of tumor immunotherapy.

8、 Yunnan University develops six fold enzyme activity nano enzyme synergistic anti-cancer

In recent years, nanoenzymes have become a research hotspot in the field of tumor microenvironment (TME) regulation and combination therapy due to their controllable catalytic activity and low-cost stable preparation advantages. However, single enzyme active materials generally have bottlenecks such as insufficient TME remodeling efficiency and a single pathway for inducing tumor death, and the therapeutic effect is far from meeting clinical needs. On May 27, 2026, Zhao Hui’s team from Yunnan University published a groundbreaking study in the top comprehensive academic journal National Science Review, reporting for the first time the Cu Zr bimetallic MOF-818 nanoenzyme with six fold natural enzyme activity, providing a new functional material platform for multi mechanism synergistic anti-tumor.

This nanoenzyme integrates six catalytic activities, including catalase and peroxidase, and can achieve efficient TME remodeling through a unique “rolling cycle catalysis” cascade effect: first, it consumes excess H ₂ O ₂ in the tumor to produce oxygen and alleviate the hypoxic microenvironment of the tumor, providing substrates for subsequent generation of reactive oxygen species (ROS), and cyclically amplifying the catalytic effect; At the same time, it can activate the prodrug acetaminophen, further inducing a large accumulation of ROS in the tumor. Unlike traditional single pathway killing strategies, MOF-818 can simultaneously activate the triple tumor cell death pathways of apoptosis, disulfide death, and copper death, effectively avoiding the problem of single pathway drug resistance and significantly improving tumor killing efficiency.

The research team further constructs functional modifications 3-MA@MOF-818 @CM Fc TRAIL nanoparticles (3MCT NPs) have been validated in tumor bearing animal models, demonstrating excellent tumor suppression effects when combined with acetaminophen, while also exhibiting good systemic biosafety. This study breaks through the limitations of traditional single function nanoenzymes and provides a new direction for the translational development of nanoenzyme based anti-tumor drugs.

9、 The team led by Zhang Chenyu from Nanjing University has published a review of extracellular microRNAs to assist in disease diagnosis and treatment

Recently, the team led by Zhang Chenyu from Nanjing University published a heavyweight review in the international authoritative journal “Immunity&Inflammation” on the field of extracellular microRNAs (miRNAs), systematically reviewing the development and core scientific issues of this field in the past 20 years, and pointing out the direction for the clinical translation of miRNAs.

In 2008, the team was the first to break the traditional understanding that miRNAs can only function inside cells and are rapidly degraded outside cells, and confirmed for the first time that miRNAs can exist stably in human serum, thus officially opening up a new direction for extracellular miRNA research. At present, the academic community has clarified that extracellular miRNAs can exist in two forms: free non vesicle encapsulated and extracellular vesicle (EV) encapsulated. Among them, EV miRNA can construct a molecular communication system across multiple tissues and organs, promoting the core cognitive upgrade of this field from “disease biomarker screening” to “cross tissue signal regulatory mediators”.

The review clearly states that EV miRNA is a precise molecular communication language for immune regulation, which can play a bidirectional regulatory role in the immune and inflammatory microenvironment, participating in core processes such as innate immune activation, adaptive immune shift, and inflammatory microenvironment remodeling. It can also serve as a biomarker to predict the efficacy of immune checkpoint inhibitors.

At present, extracellular miRNAs have achieved preliminary clinical translation in the field of early screening of tumors, but there are still challenges such as the unsolved molecular code for miRNA targeted sorting into EVs and the need to overcome bottlenecks in large-scale preparation technology. This review points out that intervention strategies based on EV miRNA have the potential to rewrite the existing diagnostic and therapeutic paradigms for cancer and autoimmune diseases, laying a theoretical foundation for the development of the next generation of novel immune therapies.

10、 A team from Shandong Medical University reveals a new anti-aging mechanism of NMN

Under the background of population aging, the research on intervention mechanisms for age-related degenerative diseases is currently a hot topic in the field of biomedicine, and the anti-aging effect mechanism of NAD+precursor NMN has always been of great concern to the industry. Recently, the team led by Mou Xiaodong from Shandong First Medical University published the latest research results in the international top journal “Autophagy” in the field of autophagy, revealing that NMN can maintain mitochondrial functional homeostasis in aging cells by activating the SIRT2 microtubule axis, elucidating a novel molecular mechanism by which NMN delays muscle aging.

Previous studies have confirmed that the NAD+level in the body decreases progressively with age, leading to a decrease in the activity of the sirtuins family of deacetylases, which is one of the core pathways driving the aging process. However, the specific molecular mechanism by which cytoplasmic localization of SIRT2 regulates aging remains unclear. This study found that in naturally aging skeletal muscle cells, NAD+depletion leads to SIRT2 inactivation, resulting in excessive acetylation of its substrate α – tubulin, disrupting microtubule dynamics homeostasis, manifested as excessive microtubule stability and abnormally weakened polymerization depolymerization kinetics; This structural defect further disrupts the dynamic repair and autophagy processes of mitochondria, leading to the accumulation of damaged mitochondria and accelerating muscle cell aging.

Research has confirmed that exogenous supplementation of NMN can activate SIRT2 by increasing cellular NAD+levels, correcting microtubule acetylation abnormalities, restoring microtubule dynamics function, thereby enhancing mitochondrial quality control and effectively alleviating the aging phenotype of muscle cells. This study deepens the scientific understanding of the NAD+pathway in regulating aging, providing a solid mechanistic basis for the development and application of NMN anti-aging, and offering new ideas for clinical intervention of age-related muscle degenerative diseases.
